THE ABC OF NAD+ from our Geoscientist π¨π½βπ¬
NAD is the most boring molecule in the body. This is because it is involved in nearly every aspect of cellular health. Without NAD we would be dead within seconds. Our cells produce NAD by piecing together precursor molecules (NMN, NR, nicotinamide) like a puzzle. When NAD is utilized/consumed, it is broken down into its individual constituents and must be pieced back together (recycled) via specific pathways (i.e. salvage pathway). Decline in NAD can be due to insufficient precursors (not enough puzzle pieces), increased consumption of NAD (i.e. through chronic inflammation), or insufficiency in recycling NAD once it has been utilized
1) NAD is an essential molecule (naturally produced in the body) that is required for mitochondrial health, energy production, regeneration and repair of cells, and the activation of multiple longevity pathways.
2) Levels of NAD within our blood and cells decline with age due to multiple physiological changes that occur with aging. This includes:
a) increased consumption of NAD to repair damaged DNA
b) chronic inflammation
c) decline in pathways critical for recycling NAD (after it has been used and broken down) to maintain intracellular pools (amount of NAD within cells)
d) Decline in physical movement, sleep, absorption of nutrients from diet, and mental health which are required from adequate NAD production
3) Supplementing with NAD precursors or engaging with compounds and lifestyle habits (i.e. fasting, resistance training) that support the functionality of pathways responsible for maintaining βyouthful levelsβ of NAD within our blood and cells has been shown to significantly reduce the risk of all-cause mortality (heart disease, cancer, autoimmune disorders, and neurodegeneration)
4) NAD levels within the body/cells have been shown to be significantly influenced Β by lifestyle habit change and supplementation. This is where viable methods for measuring NAD levels within the blood and cells are absolutely critical in order to figure out what interventions are most effective for our personalized physiology. As we age, we may need to lean heavily on supplementation because our innate abilities to maintain NAD levels begin to decline.
5) Boosting levels of NAD within the blood and cells has been shown to reverse the process of aging (i.e. rejuvenate reproductive health post-menopause), progression of chronic diseases (i.e. reverse effects or cardiovascular disease), and significantly improve both lifespan and healthspan in preclinical studies.

Longevity Drop #3π§¬π§ Interview with Dr. Nichola Conlon of NUCHIDO
1) In your research and experience, what is the most common cause for the decreasing NAD levels that accompany the aging process (i.e. not enough precursors, dysregulated pathways, chronic inflammation, etc)?
Research has shown that our NAD+ levels decline by around 50% every 20 years. NAD+ levels decline due to decreased ability of the cell to make and recycle NAD+ combined with increased demand for NAD+ by repair processes and inflammation.
NAD+ production declines with age
The cell manufactures its own NAD+ from precursors, and also by recycling the NAD+ it uses. This recycling process is called the salvage pathway. The salvage pathway is important because it is the primary way in which youthful cells make their own NAD+ and itβs the part of the system that breaks down as we age. Thatβs because the enzyme it relies on (NAMPT) declines as we get older.
NAD+ consumption increases with age
In addition to the decreased production of NAD+ there is also an increased demand for NAD+ within the cell. NAD+ is used by many different proteins within the cell and some of these proteins are known to increase with age and therefore consume more NAD+. Two of the major NAD+ consumers are PARP enzymes (repair DNA damage) and CD38 (an inflammatory protein).
PARP enzymes are involved in maintaining healthy DNA so ensuring they get enough NAD+ is vital. CD38 has been well studied and is now understood to be the largest NAD+ consumer in mammalian cells, CD38 levels increase greatly in older cells meaning it wastes a huge amount of the already limited NAD+.
Collectively, this increased demand and decreased supply leave the various cellular pathways fighting over a dwindling supply of NAD+ right at the time when they could do with more NAD+.
2) Please describe what makes your product βstand outβ from other supplements that focus on boosting NAD levels within the body.
Here at Nuchido we take a different approach to boosting NAD+ levels. We appreciate that NAD+ decline is complex and cannot be fixed by simply giving the cell more precursor materials. Nuchido TIME+ utilises systems pharmacology research which determined the precise parts of the NAD+ production network that have the biggest and most sustainable impact on boosting NAD+.
The Nuchido TIME+ supplement is the result of this research, providing the cell with a combination of ingredients that are designed to:

Prevention is always much easier than repairing damage and the same thought applies to NAD+ levels. Maintaining NAD+ levels as we age is easier than allowing them to drop significantly and then working to boost them.
As we age our NAD+ levels drop by approximately 50% every 20 years. This means that by the time we reach our 40's, the effects of reduced cellular function are starting to be felt. With declining levels of NAD+, cells cannot function like they used to. They cannot produce the right levels of energy or maintain critical cellular processes.
